Etoposide is a semisynthetic derivative of podophyllotoxin with antineoplastic properties; it interferes with the function of topoisomerase II thus inhibiting DNA synthesis, and is most active against cells in the late S and G2 phases of the cell cycle. It is a semisynthetic drug derived from the resin of the Podophyllum peltatum (mandrake root or May apple).
It is used, usually in combination with other antineoplastics, in the treatment of tumours of the testis, small cell cancer of the lung, and in acute leukaemias. It has also been tried in other solid tumours including those of the brain, gastrointestinal tract, ovary, and thymus, and some childhood neoplasms; in lymphomas, and in the treatment of Kaposi's sarcoma associated with AIDS.

Etoposide’s innovator is Novartis-Global and is known by the brand name Vepesid.

Etoposide is a cytocidal agent that is active during the G2 phase of the cell cycle. Etoposide binds to a complex of DNA and topoisomerase II. Topoisomerase II is an enzyme that coils and uncoils DNA and also examines and repairs faulty sections of DNA. Etoposide stabilizes the DNA and topoisomerase II complex and prevent further progression in the cell cycle. DNA strand breaks develop, and the cell undergoes apoptosis (programmed cell death).

Ph. Eur. 5.5: Etoposide is a white or almost white, crystalline powder. Etoposide is practically insoluble in water; slightly soluble in alcohol and in dichloromethane; sparingly soluble in methyl alcohol.

USP 30: Etoposide is a fine, white to off-white, crystalline powder. Etoposide is very slightly soluble in water; slightly soluble in alcohol, in chloroform, in dichloromethane, and in ethyl acetate; sparingly soluble in methyl alcohol.
